Genetic risk score for ovarian cancer based on chromosomal-scale length variation

چکیده

Abstract Introduction Twin studies indicate that a substantial fraction of ovarian cancers should be predictable from genetic testing. Genetic risk scores can stratify women into different classes risk. Higher treated or screened for cancer, which reduce cancer death rates. However, current do not work well. We developed score based on variations in the length chromosomes. Methods evaluated this using data collected by The Cancer Genome Atlas. synthesized dataset 414 who had serous carcinoma and 4225 no form cancer. characterized each woman 22 numbers, representing chromosome their germ line DNA. used gradient boosting machine to build classifier predict whether been diagnosed with Results chromosomal-scale variation could such highest 20% 160x (95% confidence interval 50x-450x) compared lowest 20%. we an area under curve receiver operating characteristic 0.88 0.86–0.91). Conclusion A DNA provides effective means predicting will develop

Telomere length and genetic variation in telomere maintenance genes in relation to ovarian cancer risk.

BACKGROUND Telomeres protect chromosomal ends, shorten with cellular division, and signal cellular senescence, but unchecked telomere attrition can lead to telomere dysfunction, upregulation of telomerase, and carcinogenesis. Shorter telomeres in peripheral blood leukocytes (PBL) have been associated with elevated cancer risk. Furthermore, genetic variants in and around the TERT gene have been ...

Risk of Ovarian Cancer Relapse Score

OBJECTIVE The aim of this study was to construct a prognostic index that predicts risk of relapse in women who have completed first-line treatment for ovarian cancer (OC). METHODS A database of OC cases from 2000 to 2010 was interrogated for International Federation of Gynecology and Obstetrics stage, grade and histological subtype of cancer, preoperative and posttreatment CA-125 level, prese...

Genetic variation at 9p22.2 and ovarian cancer risk for BRCA1 and BRCA2 mutation carriers.

BACKGROUND Germline mutations in the BRCA1 and BRCA2 genes are associated with increased risks of breast and ovarian cancers. Although several common variants have been associated with breast cancer susceptibility in mutation carriers, none have been associated with ovarian cancer susceptibility. A genome-wide association study recently identified an association between the rare allele of the s...

Performance of an Adipokine Pathway-Based Multilocus Genetic Risk Score for Prostate Cancer Risk Prediction

Few biomarkers are available to predict prostate cancer risk. Single nucleotide polymorphisms (SNPs) tend to have weak individual effects but, in combination, they have stronger predictive value. Adipokine pathways have been implicated in the pathogenesis. We used a candidate pathway approach to investigate 29 functional SNPs in key genes from relevant adipokine pathways in a sample of 1006 men...

Genetic variation modifies risk for neurodegeneration based on biomarker status

BACKGROUND While a great deal of work has gone into understanding the relationship between Cerebrospinal fluid (CSF) biomarkers, brain atrophy, and disease progression, less work has attempted to investigate how genetic variation modifies these relationships. The goal of this study was two-fold. First, we sought to identify high-risk vs. low-risk individuals based on their CSF tau and Aβ load a...
